SEQUEL SYSTEM

The new Sequel System is based on Single Molecule, Real-Time (SMRT) technology and delivers about 7X more reads with 1 million zero-mode waveguides (ZMWs) per SMRT Cell. The Sequel System is ideal for projects that need to generate high-quality whole genome de novo assemblies.

The PacBio RS II is suitable for whole genome sequencing of smaller organisms and targeted sequencing of DNA and RNA.

Advantages of PacBio technology

  • Really long reads

    This technology achieves read lengths up to 30,000 bp with a mean size of 4,200 to 8,500 bp.

  • Near-random distribution of errors

    This is a great advantage because increasing the coverage you can correct the errors while in the case of non-random errors even with a huge increasing of the coverage the error rate can not be corrected in the regions prone to errors.

  • No PCR bias

    Because the library preparation has not any PCR step the DNA that is sequenced is the native DNA from the sample.

  • The capability of directly detecting modified nucleotides

    This is currently the only technology able to directly detect all the types of nucleotide modifications. A really high coverage is needed for this detection.

  • Circular Consensus Reads (CCS)

    Pacbio offers two kinds of reads: Standard Long reads and CCS (Circular Consensus Sequencing) reads. CCS reads have a low error rate and a length sufficient to solve many long repeats in genomes. In many projects can be useful to join the possibilities that supply the long standard Pacbio reads with the accuracy of CCS reads.
